The short version of casein vs whey: whey is the faster protein and the better bet around training, casein is the slower one and the better bet before a long gap without food. Both build muscle. Neither builds much of it if your daily protein total is short, and the difference between them is smaller than the difference between hitting your total and missing it.

That answer disappoints people who came looking for a winner. The supplement aisle has spent thirty years selling these two as rival philosophies, when they're simply the two halves that separate out when milk curdles. Whey is the liquid. Casein is the curd. About 80% of the protein in milk is casein and about 20% is whey, which means a plain glass of milk is already the blend most people end up buying separately.

What Is the Actual Difference Between Casein and Whey?

Both are complete proteins with all nine essential amino acids and both are highly digestible. The separation happens in your stomach. Casein is acid-sensitive, so it clots into a soft gel that breaks down slowly. Whey stays soluble and empties out of the stomach fast.

How Fast Does Each One Actually Digest?

The foundational experiment is still the cleanest. Boirie et al. (1997), writing in the Proceedings of the National Academy of Sciences, fed healthy adults a single meal of carbon-13-labelled casein or whey and followed the leucine through the body. Whey produced what the authors called a dramatic but short increase in plasma amino acids. Casein produced a prolonged plateau of moderate hyperaminoacidemia, which they attributed to slow gastric emptying.

The downstream effects split too. Whole-body protein synthesis rose 68% after whey and 31% after casein. Protein breakdown was inhibited 34% after casein and not at all after whey. Two different tools, in other words. Whey pushes the build rate up. Casein mostly stops the teardown.

That kinetic difference is why whey wins acute muscle protein synthesis comparisons. Pennings et al. (2011), in the American Journal of Clinical Nutrition, gave 48 older men (mean age 74) a 20g dose of intrinsically labelled whey, casein or casein hydrolysate and measured muscle fractional synthetic rates directly. Whey came out at 0.15%/h, casein hydrolysate at 0.10%/h, and intact casein at 0.08%/h. The authors credited both the faster absorption and the higher leucine content.

Does the Amino Acid Profile Matter, or Just the Speed?

Both, and they are hard to separate. Whey is the more leucine-dense of the two, at roughly 10 to 11% of its amino acids against about 8% for casein, and leucine is the amino acid that flips the molecular switch for muscle protein synthesis. The International Society of Sports Nutrition position stand on protein and exercise (Jager et al., 2017) concluded that rapidly digested proteins high in essential amino acids and adequate in leucine are the most effective at stimulating synthesis. That sentence is basically a description of whey.

You can see the two factors pull apart in Tang et al. (2009) in the Journal of Applied Physiology. The researchers matched three drinks for essential amino acid content (10g each) and gave them to young men after unilateral leg resistance exercise: whey hydrolysate, micellar casein, or soy isolate. Whey and soy both beat casein at rest and after exercise, which is the speed effect. Whey then beat soy after exercise as well, despite both being fast, which is the leucine effect.

The practical version is the leucine threshold. A feeding needs somewhere around 2 to 3 grams of leucine to maximally switch on synthesis in a young adult, and more than that in an older one. A 25g scoop of whey clears it comfortably. A 25g scoop of casein sits closer to the line, which is one reason the casein trials tend to use 30g to 40g rather than 20g.

What Happens Over Weeks and Months?

Acute synthesis rates are a proxy, not an outcome. Plenty of things raise synthesis for three hours and change nothing after twelve weeks. So the question that matters is whether whey's head start survives a real training block.

It mostly does not, or at least not by much. Li et al. (2026) ran a Bayesian network meta-analysis in Frontiers in Nutrition across 36 randomized trials, 89 trial arms and 1,552 participants. Whey ranked first for lean body mass (SUCRA 0.91, mean difference +1.24 kg versus placebo, 95% credible interval 0.61 to 1.87). Milk came second at +1.18 kg. Casein came third at +0.92 kg. For strength, whey ranked first again with casein a short way behind. The ordering is consistent and the margins are small, with intervals that overlap.

Age moves the ranking. In adults 65 and over, whey's advantage narrowed and casein's ranking improved, which fits the pattern that older muscle responds less sharply to a fast amino acid spike and may do better with a longer signal.

The bigger context comes from Morton et al. (2018) in the British Journal of Sports Medicine, a meta-analysis of 49 studies and 1,863 participants. Protein supplementation added 0.30 kg of fat-free mass and 2.49 kg to one-rep max strength over training alone. And the effect flatlined once total intake passed about 1.62g per kg of bodyweight per day. Once you're at your total, the source is a rounding error.

One trial cuts against the grain and deserves an honest mention. Demling and DeSanti (2000), in Annals of Nutrition and Metabolism, put overweight police officers on a 12-week hypocaloric diet at 80% of predicted needs with resistance training and 1.5g/kg/day of either a casein hydrolysate or a whey hydrolysate. Everyone lost about 2.5 kg of bodyweight. The casein group lost 7.0 kg of fat and gained 4 kg of lean mass, the whey group lost 4.2 kg of fat and gained 2 kg, and strength rose 59% in the casein group against 29% in the whey group. Those numbers are extraordinary, the groups were small at 14 each, hydrolysates are not what most people buy, and nobody has replicated it in 25 years. Treat it as an interesting outlier rather than a reason to reorganize your shelf.

The One Job Casein Clearly Owns: Before Sleep

Sleep is the longest stretch of the day with no protein coming in. A group at Maastricht University spent a decade asking whether you can do anything useful with it, and the answer turned out to be yes.

Res et al. (2012) had 16 young men train in the evening, eat a standard recovery meal, then take 40g of casein or a placebo 30 minutes before bed. Overnight muscle protein synthesis was about 22% higher in the casein group, and whole-body protein balance flipped from negative to positive. Snijders et al. (2015) then ran the 12-week version in the Journal of Nutrition with 44 young men on a supervised program. The pre-sleep casein group added significantly more quadriceps cross-sectional area than the placebo group and more one-rep max strength, with no difference in fat mass.

A 2021 systematic review by Reis et al. in the Journal of Science and Medicine in Sport pooled nine articles and concluded that 20g to 40g of casein roughly half an hour before sleep raises overnight whole-body protein synthesis in both young and older men, while the chronic body-composition effects are more modest. Whey cannot do this job. A bedtime whey shake is absorbed and gone inside two hours, leaving the other six in the same fasted state you would have been in anyway. This is the single clearest use case either protein has.

Does Either One Speed Up Recovery?

Less than the marketing suggests. Davies et al. (2018), in Nutrients, pooled eight studies and 13 randomized trials on whey and the recovery of muscle function after resistance training. They found positive small to medium effects that stayed consistent across the hours and days after training, and called for more comprehensive work before anyone gets confident. Small and real, in other words, not transformative.

Casein's contribution to recovery is mostly the overnight one already described, plus its effect on protein breakdown. Neither protein is a recovery supplement in the way sleep, total food intake and sane training volume are. If soreness or dead-legged sessions are your actual complaint, the lever is usually elsewhere.

What About Price and Practical Use?

Whey concentrate is normally the cheapest protein per gram on the shelf, because it's the least processed form of the more abundant by-product. Whey isolate costs more for a slightly higher protein percentage and much less lactose. Micellar casein usually sits above whey concentrate, partly because demand is lower and partly because the slow-release positioning carries a premium.

You can skip the second tub entirely. Cottage cheese, Greek yogurt and plain milk are all predominantly casein, which means a bowl of cottage cheese before bed is doing the same job as a casein shake with none of the chalky texture. Milk-protein blends, sold as such, land in the middle by design.

One texture warning worth having in advance: 40g of casein in a normal amount of water sets into something closer to pudding than a drink. Use more liquid than you think, or eat it with a spoon and stop pretending it is a shake.

Two things matter more than any of this. Your daily total, and whether it's spread across the day rather than dumped into dinner. The Bottom Line

Casein vs whey is a timing question, not a quality question. Whey digests fast, carries more leucine, and produces the bigger acute muscle protein synthesis response. Casein digests slowly, blunts protein breakdown, and is the only one of the two with a real evidence base for the overnight window. Across whole training blocks the measured difference in lean mass is a few hundred grams, with overlapping confidence intervals.

Practical rule: whey around training and at meals, casein before sleep or before a long gap without food, and a blend if you only want one tub. Then stop optimizing the source and go fix the total.

The reason this debate has lasted so long is that it's a satisfying thing to argue about and a cheap thing to change. Swapping tubs feels like progress.
